Additive Effects
Researchers have found that dipyridamole and ASA have separate, independent, antithrombotic effects. The dosage ratio of aspirin to extended-release dipyridamole in Aggrenox®/Asasantin® was shown ex vivo to provide additive inhibition of platelet aggregation. This dosage ratio was used in ESPS 2, the 6602-patient study that showed that the formulation in Aggrenox®/Asasantin® reduced risk of stroke by 37% vs. placebo in patients with prior ischaemic stroke or TIA, and nearly doubled the risk reduction of ASA alone (50 mg/day). Each enteric-coated granule of extended-release dipyridamole has an acidic core designed to promote absorption.
