Endothelium
Dipyridamole has effects on both platelets and the endothelium. Dipyridamole inhibits cell uptake of adenosine, which increases intraplatelet cAMP, an inhibitor of platelet aggregation.
Dipyridamole also inhibits breakdown of cyclic GMP, which also inhibits platelet aggregation.
Unlike the prolonged effect of ASA, the effects of dipyridamole are shorter, requiring either multiple daily dosing or more prolonged plasma concentrations.
